El 26 de octubre de 1949 el reportero Gabriel García Márquez fue enviado al antiguo convento de Santa Clara, que iba a ser demolido para edificar sobre él un hotel de cinco estrellas, a presenciar el vaciado de las criptas funerarias y a cubrir la noticia. Se exhumaron los restos de un virrey del Perú y su amante secreta, un obispo, varias abadesas, un bachiller de artes y una marquesa.

Al pueblo ha llegado «la mala hora» de los campesinos, la hora de la desgracia. La comarca ha sido «pacificada» después de tanta guerra civil. Han ganado los conservadores, que se dedican a perseguir cruel y pertinazmente a sus adversarios liberales. Al alba de una mañana, mientras el padre Ángel se dispone a celebrar la misa, suena un disparo en el pueblo. Un comerciante de ganado, advertido de la infidelidad de su mujer por un pasquín pegado a la puerta de su casa, acaba de matar al presunto amante de ésta.

En Macondo transcurre la historia de un entierro imposible. Ha muerto un personaje extraño, un antiguo médico odiado por el pueblo, y un viejo coronel retirado, para cumplir una promesa, se ha empeñado en enterrarle ante la oposición de todo el pueblo y sus autoridades. Como en una tragedia griega -el libro lleva como lema una cita de Antígona que recuerda la prohibición de enterrar el cuerpo del Polinices-, el viejo coronel, su hija y su nieto van a cumplir la ominosa tarea.

En Barcelona, una prostituta que va entrando en la vejez entrena a su perro para llorar ante la tumba que ha escogido para sí misma. En Viena, una mujer se vale de su don de ver el futuro para convertirse en la adivina de una familia rica. En Ginebra, el conductor de una ambulancia y su esposa acogen al abandonado y aparentemente moribundo ex presidente de un país caribeño, solo para descubrir que sus ambiciones políticas siguen intactas.
