Monte Carlo, Mai 1904: Prof. van Dusen und sein Begleiter Hutchinson Hatch machen auf ihrer Weltreise Halt im monegassischen Spielerparadies. Hier treffen sie den rumänischen Hochstapler Manolescu, der als angeblicher Marchese delle Rocca ungewöhnlich viel Glück am Roulette-Tisch hat und Unsummen Bargeld einstreicht. Im Vertrauen teilt er Prof. van Dusen und Hatch mit, dass er nach System gewinnen würde und am morgigen Tag seinen Trick verraten könne. Doch so weit kommt es nicht, denn in der Nacht wird Manolescu in seinem verschlossenen Hotelzimmer erschossen und das gewonnene Geld ist weg. Professor van Dusen macht sich auf die Suche nach dem Mörder, der in einem hermetisch verschlossenen Raum getötet hat.
